Primers: to make 5 uL of 5 µM (pmol/µl) in 5uL dillution, mix 2.5uL of our 10x primer stock with 2.5uL water.
Template: in most cases, one well will contain 1.75uL of cleaned-up reaction outcome + 8.25uL of water = 10uL
for Pre-Mixed sequencing: combine the above for 15uL sample in each well
